Unless your story follows the “love at first sight” trope in romance, there’s no hurry in building chemistry between two characters. Take your time and make sure that every moment the two spend together counts.

1. Let Them Reveal Themselves to Each Other
Genuine reactions form the core of chemistry. Thus, create enough scenes for the two characters in question to know each other, so that when they choose each other, whether as enemies or friends, the audience can relate to their decision.
At the same time, character exposition allows your actors to develop their characters more intimately, which helps with chemistry.
2. Strong Camaraderie or Strong Conflicts—You Decide
While the characters and the relationship between them are most definitely allowed to transform over the course of the story, you should pick a solid ground (friendship or animosity) to start their association.
Either they share strong similarities, or are polar opposites when it comes to their life ideals.
Once you have that decided, double down on the emotion with scenes that explore this aspect of their relationship. Banter is a tried and tested way to spark chemistry between two characters—romantically associated or not.

4. Stick to Conflict That Is Coherent with the Themes
Always try to align the interactions between two characters with the story's themes.
This is because misplaced character interactions can throw off audiences and kill chemistry as a whole if these scenes do not contribute effectively to the story.

Kim and Jimmy are my most beloved couple from recent TV. Their chemistry lies in the way they understand and protect each other.
Throughout the series, we see various instances of both conflict and harmony between them, as Jimmy bends the rules to earn money or prove himself a worthy lawyer. After a traumatic death, Kim breaks up with him, realizing that her support is enabling him to lead a life of crime.
Their chemistry feels so real because, despite opposing ideals, they share a lot of similarities—both are self-made, have the right intentions at heart despite their own twisted ways, have been betrayed by people they were loyal to, and finally, have nobody but each other in their lives.
4. Harry, Ron, and Hermione: Harry Potter

Hermione becomes a subtle bridge between Harry and Ron, and between Harry and the magical world, uniting them despite their differences, while she grows through her adventures with the boys.
By the end of the franchise, they embody harmony amidst adversity, and that is what makes their chemistry so memorable. Once again, their relationship blossoms over time, which allows the audience to connect with them.

However, alongside the strong emotional scenes in the present, when Ilsa meets Rick at his cafe, accompanied by her husband, seeking transit visas to America, it is both the actors’ charismatic yet restrained performances that create an electrifying chemistry between them.
The way Bergman’s Ilsa looks at Bogart’s Rick when she sees him for the first time, after an entire year since she abandoned him, and requests Sam to play their song—pure chills!
